HFE-TfR complex
HFE is a class I MHC-related protein that is mutated in the iron overload disease hereditary hemochromatosis (HH). HH is more common than cystic fibrosis, muscular dystrophy and phenylketonuria combined. HFE binds to the transferrin receptor (TfR), a recptor by which cells acquire iron-loaded transferrin . We have determined the 2.8 Å crystal structure of a complex between the extracellular portions of HFE and TfR.
